Effective, homogeneous and transient interference with cytosine methylation in plant genomic DNA by zebularine

چکیده
Covalent modification by methylation of cytosine residues represents an important epigenetic hallmark. While sequence analysis after bisulphite conversion allows correlative analyses with single-base resolution, functional analysis by interference with DNA methylation is less precise, due to the complexity of methylation enzymes and their targets. A cytidine analogue, 5-azacytidine, is frequently used as an inhibitor of DNA methyltransferases, but its rapid degradation in aqueous solution is problematic for culture periods of longer than a few hours. Application of zebularine, a more stable cytidine analogue with a similar mode of action that is successfully used as a methylation inhibitor in Neurospora and mammalian tumour cell lines, can significantly reduce DNA methylation in plants in a dose-dependent and transient manner independent of sequence context. Demethylation is connected with transcriptional reactivation and partial decondensation of heterochromatin. Zebularine represents a promising new and versatile tool for investigating the role of DNA methylation in plants with regard to transcriptional control, maintenance and formation of (hetero-) chromatin.

Involvement of Cytosine DNA methylation in different developmental stages of Aeluropus littoralis
DNA methylation as epigenetic mark plays a key role in normal differential and developmental processes as well as in dynamic gene regulation at the genomic level. To assess DNA methylation pattern in different developmental stages of Aeluropus littoralis, methylation sensitive amplified polymorphism (MSAP) was used. Methylation and demethylation status at the CCGG recognition site were...

DNA methylation as an epigenetic mediator plays the important role in spatial and temporal gene regulation and ensures the stability and the plasticity of organism. In this investigation, methylation sensitive amplification polymorphism (MSAP) were assessed in CCGG sites on a halophytic plant, Aeluropuslittoralis in response to different temperature stresses including freezing...
متن کاملA Comparative Analysis of 5-Azacytidine- and Zebularine-Induced DNA Demethylation
The nonmethylable cytosine analogs, 5-azacytidine and zebularine, are widely used to inhibit DNA methyltransferase activity and reduce genomic DNA methylation. In this study, whole-genome bisulfite sequencing is used to construct maps of DNA methylation with single base pair resolution in Arabidopsis thaliana seedlings treated with each demethylating agent. We find that both inhibitor treatment...
متن کاملThe plant genome's methylation status and response to stress: implications for plant improvement.
Plant improvement depends on generating phenotypic variation and selecting for characteristics that are heritable. Classical genetics and early molecular genetics studies on single genes showed that differences in chromatin structure, especially cytosine methylation, can contribute to heritable phenotypic variation. Recent molecular genetic and genomic studies have revealed a new importance of ...
